TESTRÖ-QUANT 3 S (TQ 3 S)

TQ 3S serves for quantifying bacteria, moulds and yeasts, and hygienically relevant bacteria, in water soluble cooling lubricants and other liquids.

1. Characterisation
TESTRÖ-Quant 3S is a threepart medium that is attached to a lid. This lid seals a little tube, in which the mediums are protected against dehydration and contamination.

1. Medium for determining the total bacteria count.
2. Pink medium for coliform bacteria.
3. Medium for moulds and yeasts.

On medium 1 colonies of predominantly bacteria cultivate after 2-3 days, on medium 2 colonies of yeasts and moulds only. On the medium for hygienically relevant bacteria only red coloured colonies are to be analysed. The cultivation of moulds and yeasts and colourless colonies of bacteria on the pink medium is possible. The analysis, however, is not affected by them.

2. Implementaion of the meduim analysis

Fill in the sticker and attach it to the tube.

Unscrew the lid directly above the sample drawing and withdraw the medium without touching the coated sides.

Dip the medium immediately into the liquid you wish to analyse, or into the sample inside the receptacle (5-10s), so that the coatings become completely moistened. The medium is to be dipped into undisturbed liquids only. See that floating oil and other foreign matters are removed before the dipping. TQ 3S is to be dipped into the coolant for at least 10 seconds. Only in this case can correct and comparable values be obtained. This is significant for all mediums.

Let the medium drip, then dab off the lower edge in upright position onto a clean filter paper.

In case you used the receptacle for the sampling, it has to be cleaned now. Then insert the medium into the receptacle and screw it tight carefully.

Put TESTRÖ-Quant 3S in upright position into an incubator that is adjusted to 34-37°C.

After 16-24 hours the time of incubation is over. Analyse the mediums according to the analogue pictures.

Without diluting the sample the method is suitable for determining colony numbers up to 106/ml. In case you want to determine colony numbers higher than 106/ml, the sample has to be watered down. Proceed as follows: Fill 100ml, or 1000ml respectively, tap water into a clean, repeatedly carefully rinsed and desiccated, sealable bottle. The water should have drinking water quality; diluting water with more than 100 bacteria per ml is not suitable. the suitability can be checked with TESTRÖ-Quant 3S. Let the water run for 5 minutes or boil it for 15 minutes and let it cool down afterwards, before filling the bottle. Then add 1 ml of the liquid you wish to analyse with a clean pipette (e.g. non-returnable pipette). After sealing the bottle shake the mixture well for a minute. There has to be enough space in the bottle, so that by shaking a good stirring is ensued. Then dip the medium into the dilution and proceed as is described above (2.1-2.7). The dilution factor is to be considered when analysing the dilution. If, for example, the sample diluted with 100ml shows 106 bacteria or yeast cells after incubation and the true value for the dilution 1:100 is 108 bacteria or yeast cells per ml.

Only the amount of the single colonies but not their size is relevant for the analysis. In case of a very high colony count a bacterial carpet has been cultivated, i.e. between the colonies there are no spaces any more. The whole surface is then abundantly covered and therefore shines. In this case the sample can be watered down for determining the colony count (see 2.8). Universal limiting values that justify the application of preservatives cannot be denounced but ensue by experience.

3. Analysis
For reading of the result the TQ 3S can be taken from the receptacle, if this should have got steamed up. Bacteria and fungi do not have legs and therefore do not jump away from the medium. You must not touch the colonies, in any case unnecessary.

3.1 Total bacteria (see medium 1)
The determination of the total bacteria ensues with the aid of the comparison pictures, i.e. the colonies are not counted.

3.2 Coliform bacteria (medium 2)
The pink-coloured part of the divided parts shows proof of coliform bacteria. Coliform bacteria cultivate strikingly red coloured colonies and are therefore well distinguishable from other – colourless – colonies that also cultivate on this side.

3.3 Fungi (medium 3)
On the yellowish part of the divided side moulds cultivate woolly colonies and yeasts round white- or red-coloured, mat colonies that sometimes have runners.

3.3.1 Moulds
Moulds cultivate their colonies from single spores, filament particles or filament aggregates. The congruence with the analogy pictures (pic. 3) therefore represents only approximated values.

3.3.2 Yeasts
Yeasts cultivate colonies similarly to bacteria and can be analysed like them. (pic. 1)

Disposal of used culture mediums
On the incubated mediums are cultures of microorganisms. They must not be touched. After analysing them, the mediums have to be destroyed.The killing of the microorganisms can ensue by burning the receptacles together with the mediums depositing or by depositing the mediums and receptacles into an aggressive disinfectant. In the latter case the receptacles must be unscrewed and both the parts deposited into the disinfectant over night.

Storage
TESTRÖ-Quant 3S that have not been opened can be preserved for up to 6 months when stored in a cool and dark place. When storing the TQ 3S in the refrigerator they can be preserved for approximately one month longer (no guarantee).
